Dear Colleagues, this week we will examine Thymol together, a phenolic antiseptic substance with antifungal activity included in the Spanish National Formulary.<\/p>\n

Formula;<\/strong><\/p>\n

Rp.<\/strong><\/p>\n

Thymol                        100 mg<\/strong><\/p>\n

Alcohol 960    <\/sup>               2 g<\/strong><\/p>\n

Glycerin                      2 g<\/strong><\/p>\n

Distilled water q.s.   100 g<\/strong><\/p>\n

Substances in the formula;<\/strong><\/p>\n

Isopropyl metacresol – Thymic acid – Thymol – Thymolum – Timolo [FU];<\/strong><\/p>\n

Thymol occurs as colorless, large crystals or crystalline powder with a pungent thyme odor. According to the British Pharmacopoeia (BP), its melting point is between 48-52\u00b0C. Its solubilities according to the BP: Very slightly soluble in water; freely soluble in alcohol and ether; freely soluble in volatile and fixed oils; sparingly soluble in glycerol; soluble in dilute solutions of alkali hydroxides. Thymol should be stored in airtight containers. It should be protected from light.<\/p>\n

Although it is more effective than phenol, its use is limited due to its low solubility in water, higher irritancy, and reactivity toward proteins. Thymol is used as a deodorant in mouthwashes and gargles. Furthermore, it is beneficial topically in certain skin diseases due to its counterirritant and antiseptic effects. Thymol iodide is also utilized in the treatment of skin diseases. Thymol is included alongside other volatile substances in the composition of preparations used for colds, coughs, and various respiratory tract diseases. It is added as an antioxidant to halothane, trichloroethylene, and tetrachloroethylene at a concentration of 0.01%.<\/p>\n

Thymol exerts its antiseptic and disinfectant action by increasing the permeability of the cytoplasmic membrane of bacteria and fungi. Thus, it exhibits bactericidal and fungicidal effects. Thymol is applied topically.<\/p>\n

Glycerol – Glycerine – Glycerol 85%;<\/strong><\/p>\n

It is used for its solvent, vehicle, preservative, and viscosity-enhancing properties. It is an important moisturizing agent. Glycerin is a clear, colorless or very pale, hygroscopic, syrupy liquid that is odorless or nearly odorless, with a sweet taste. It is miscible with water and alcohol; slightly soluble in acetone; practically insoluble in chloroform, ether, fixed oils, and essential oils. The pH of its aqueous solutions is neutral to litmus. According to the United States Pharmacopeia (USP), the pH of Glycerin Oral Solution, USP is between 5.5-7.5. Strong oxidizing agents form explosive mixtures with glycerin.<\/p>\n

Preparation of the formula;<\/strong><\/p>\n

Thymol is dissolved in alcohol, and glycerin is added. Finally, distilled water is added. To obtain a clear solution, approximately 1% Tween 20 should be added, and it should be stored in an amber glass bottle.<\/p>\n

This solution is known as aqueous thymol.<\/p>\n

Formula for treating onychomycosis cases;<\/strong><\/p>\n

Chloroform q.s.    100 g<\/strong><\/p>\n

Preparation of the formula;<\/strong><\/p>\n

Thymol is dissolved in chloroform and stored in an amber glass bottle. This formula is used to treat onychomycosis cases.<\/p>\n
