COLLUTORIUM METHYLENI COERULEI – Methylene Blue Collutory<\/strong>Dear Colleagues, this week I will share with you a formula used as an antiseptic for aphthae. Until recently, difficulties have been experienced in procuring this commercial preparation, which contained the active substances found in this formulation and was available on our pharmacy shelves under the name Buco Bleu Collutory. This preparation, which we can easily prepare in our laboratories, is of great importance in terms of providing a compounded solution for our patients’ treatments.Collutory<\/strong>If we recall the definition; they are pharmaceutical dosage forms in solution used in oral hygiene and the treatment of diseases affecting the oral mucosa, including the gums. To enhance their efficacy and prolong their duration of action, they may also contain thickening agents such as glycerin, honey, or syrup as vehicles. They frequently contain antiseptic substances (such as methylene blue and iodine compounds) as active ingredients.<\/p>\n

Among the preparation’s active ingredients, methylene blue has an antiseptic effect, while resorcinol exhibits an antifungal effect. It is used as an oral antiseptic. There are no known contraindications for this formula in the form of an externally applied solution. It is applied to the affected mucosa or skin 3 times a day using a brush.<\/p>\n

After application, the patient should not spit or swallow the medicine. The formula can also be used as a gargle after adding 8\u201310 drops to a glass of water.Returning to our formula;<\/strong><\/p>\n

Substances in the formula<\/strong><\/p>\n

Methylthionine chloride- Bleu de methylene- Methylene blue- Methylenii caeruleum- Methylthioniniumchlorid- Tetramethylthioninchlorid;<\/strong>Methylene blue is found in the form of dark green, odorless or very slightly scented crystals or crystalline powder. Its solutions in water and alcohol are dark blue. It dissolves in water at a ratio of 1:25 and in alcohol at a ratio of 1:65. It is soluble in chloroform. According to the United States Pharmacopeia (USP), the pH of its aqueous solution is between 3.0 and 4.5. Methylene blue is a water-soluble thiazine dye. It is used orally as a urinary tract antiseptic and parenterally as an antidote in the treatment of cyanide poisoning or drug-induced methemoglobinemia (amyl nitrite and sodium nitrite are preferred since its efficacy in cyanide poisoning is weak). Due to its antiseptic effect, it is also included in the formulation of some antiseptic preparations applied to the eyes, mouth, pharynx, and skin.Dibromsalicylii- Resorcine- Dioxybenzol- Resorcinol;<\/strong>It is a white crystalline substance with a characteristic odor. Its color turns red under the influence of air and light. It has a pungent taste. It is soluble in 1 part water and 1 part alcohol, as well as in ether, glycerin, and benzene, while it is insoluble in chloroform. <\/strong>It is incompatible with acetanilide, camphor, chloral hydrate, menthol, and phenol. Resorcinol is used as an antiseptic and disinfectant. It is used externally (ointment 10-20%, solution 0.1%) for psoriasis, acne, seborrhea, eczema, and other skin diseases. Application to very large surfaces should be avoided. M.D. is 0.50-1.5 g.Spiritus anisi- Spirit of anise- Anise tincture;<\/strong>Anise, containing essential oil (cineole, d-terpineol, limonene) obtained from the fruits of Illicium verum and possessing stimulant, stomachic, antispasmodic, carminative, and diuretic effects, is used in this formula for flavoring purposes.Preparation <\/strong>

Ammonium chloride
\n3.0 g<\/p>\n

Ethanol 96% (V \/ V)
\n40.0 g<\/p>\n

Purified water
\n55.0 g<\/p>\n<\/table>\n

Purified water is added to the aniseed oil with continuous shaking, dissolved in 96% ethanol, and then ammonium chloride is added. If the liquid contains particulate matter, it is shaken with 3 g of talc, left to stand for a few hours with occasional stirring, and then filtered.Glycerol- Glycerine- Glycerol-85 %;<\/strong>Glycerin is a clear, colorless or very pale, hygroscopic, syrupy, odorless or faintly scented liquid with a sweet taste. It is miscible with water and alcohol; slightly soluble in acetone; practically insoluble in chloroform, ether, fixed oils, and essential oils. The pH of its aqueous solutions is neutral to litmus. According to the United States Pharmacopeia (USP), the pH of Glycerin Oral Solution, USP is between 5.5 and 7.5. Strong oxidizing agents form explosive mixtures with glycerin.Aqua- Water- Water- Wasser- Eau;<\/strong>In pharmacy, water refers to distilled water. Demineralized water is also used today. Water that has been distilled twice is called double-distilled water.Preparation of the formula<\/strong>The calculated amount of resorcinol is dissolved in 14 ml of distilled water. Methylene blue is wetted with the remaining 6 ml of distilled water. Glycerin is added and dissolved with gentle heating (below 600 <\/sup>\u00b0C). After cooling, the resorcinol dissolved in distilled water and the anise syrup are added.<\/p>\n
