Substances that enable the separation or shedding of the horny layer of the epidermis are called keratolytic agents or substances that induce keratolysis. Formula:

Rp.

Salicylic acid     6.00 g

Resorcinol       6.00 g

Lanolin             5.00 g

Vaseline             5.00 g

Ingredients in the formula:

Salicylic acid- Acidum salicylicum- Salicylsäure- Acide salicylique;

It consists of white crystals. It is soluble in 500 parts water, 15 parts boiling water, 27 parts alcohol (99%), 2 parts ether, 80 parts olive oil, 100 parts glycerin, and 55 parts chloroform. Salicylic acid accelerates desquamation. It exhibits a mild fungicidal effect. Salicylic acid is a beta-hydroxy acid (BHA) with the chemical formula C6H4(OH)CO2H. This colorless, crystalline organic acid is generally referred to as a plant hormone. It is a product of salicin metabolism. It possesses chemical properties similar to acetylsalicylic acid, known as aspirin. It is used in the treatment of fungal species such as tinea barbae, tinea capitis, and tinea cruris. It is included in formulations due to its sufficiently potent analgesic and antipyretic, and rarely antiseptic effects, and is used externally as an analgesic in anti-rheumatic applications. It is preferred for its caustic effect against mycotic eczema. In dermatology, salicylic acid is often used in combination with lactic acid for the treatment of warts and corns. The mechanism of action of salicylic acid and lactic acid in wart treatment is not completely understood. It is thought to be related to the mechanical removal of wart virus-infected epidermal cells as a result of their keratolytic effects. They are also effective against corns through their keratolytic activities. It is used as a gargle in the treatment of tonsillitis. It is used at a dose of 0.50% against fermentation in foods such as beer, wine, meat, etc.

Resorcinol- Dibromsalicylii-  Resorcine-  Dioxybenzene- Resorcin;

It is a white crystalline substance with a characteristic odor. Its color turns reddish under the influence of air and light. It has an acrid taste. It is soluble in 1 part water and 1 part alcohol, as well as in ether, glycerin, and benzene, but insoluble in chloroform. It is incompatible with acetanilide, camphor, chloral hydrate, menthol, and phenol. Today, resorcinol is used externally as an antiseptic and disinfectant. It is applied externally (ointment 10-20%, solution 0.1%) in psoriasis, acne, seborrhea, eczema, and other skin diseases. Application to very large surface areas should be avoided. M.D. is 0.50-1.5 g.

Lanolin- Wool fat- Adeps lanae- Wool fat- Lanolin anhydrous:

Lanolin is an ointment base. It is widely used in topical pharmaceutical formulations and cosmetic products. It is used in the preparation of water/oil type creams and ointments. When mixed with suitable vegetable oils or vaseline, creams with emollient properties that penetrate the skin are obtained. Due to this property, it facilitates the absorption of drugs. Lanolin blends with approximately twice its weight of water without phase separation, producing stable emulsions. Lanolin is a purified waxy substance obtained from the wool of sheep (Ovis aries, Bovidae). It mainly consists of fatty acid esters of cholesterol, lanosterol, and fatty alcohols. Its most important property is its capacity to absorb up to 30% water. It is a pale yellow, greasy substance with a characteristic odor. When melted, it turns into a clear or almost clear yellow liquid. According to the United States Pharmacopeia (USP), its melting point is between 38-44°C. The USP allows it to contain up to 200 ppm of a suitable antioxidant. 10 g of lanolin must retain at least 20 ml of water. It is practically insoluble in water; sparingly soluble in cold alcohol, freely soluble in boiling alcohol. It is easily soluble in benzene, chloroform, ether, and petroleum ether. It may slowly undergo auto-oxidation during storage. To prevent this, the addition of butylated hydroxytoluene (BHT) is permitted. In case of prolonged heating, its color darkens and an unpleasant odor may develop.

Vaseline- White petrolatum- White soft paraffin- Officinal petrolatum- Paraffin, soft, white [BP; P.Cx.79]- Petrolatum, white [NF; USP]- Vaselin, weisses [PH; Ph.Eur.];

Vaseline is used for the treatment of skin irritations and for skin care. Specially processed vaseline-containing preparations are also used for wound care and wound dressings. Vaseline is a greasy substance that provides a soothing effect by softening the skin, makes its surface smooth, and is effective against disorders causing dryness or scaling. It provides a moisturizing effect on the skin, thereby helping to revitalize it. Vaseline (white soft paraffin) is obtained by bleaching yellow soft paraffin. According to the United States Pharmacopeia (USP), a suitable stabilizer may be added to its composition. Vaseline is incorporated into the composition of topical pharmaceutical preparations as an emollient ointment base. It is not easily absorbed through the skin. It is also used in creams, transdermal preparations, and cosmetic products. Hypersensitivity reactions associated with its topical use have been reported. Polycyclic aromatic hydrocarbon impurities it carries may cause allergic reactions. The amount of these impurities depends on the source of the vaseline and its degree of purification. Hypersensitivity reactions caused by white vaseline are fewer than those caused by yellow vaseline. For this reason, white vaseline is preferred in cosmetic products and pharmaceutical preparations.

Yellow vaseline is derived from petroleum. It is a purified mixture of semi-solid saturated hydrocarbons with the general formula CnH2n+2. Yellow vaseline is a pale yellow or yellow, semi-transparent mass with a soft consistency. It is odorless and tasteless. It is practically insoluble in acetone, alcohol, glycerin, and water; soluble in benzene, carbon disulfide, chloroform, ether, hexane, and numerous fixed and volatile oils. The United States Pharmacopeia (USP) allows the addition of a suitable stabilizer to its composition. It should be stored in well-closed containers in a cool and dry place, protected from light. Vaseline is quite a stable substance. However, it may cause stability problems due to the very small amounts of impurities it carries. These impurities can be oxidized under the influence of light to cause discoloration, resulting in unwanted odors. Such oxidations can be prevented by the addition of an antioxidant such as butylated hydroxyanisole, butylated hydroxytoluene, or alpha-tocopherol. It should not be heated for prolonged periods above the temperature required to make it completely fluid (approximately 70°C).

Preparation of the formula:

There is an incompatibility between resorcinol and salicylic acid that causes discoloration upon direct contact. To prevent this incompatibility, a few grams of water must be added to dissolve the resorcinol, while on the other hand, the salicylic acid must be levigated with vaseline in a mortar.

Dissolve the resorcinol in 5 g of water in a beaker. In a mortar, levigate the salicylic acid with the vaseline softened by gentle warming until a homogeneous mixture is formed. Add the lanolin, also softened by gentle warming, to the mixture and incorporate. Finally, add the resorcinol solution dissolved in water gradually to the mixture to complete the formula.

Wishing you a good week…

Spec. Pharm.
